734 Development of multicellular organoids for skin injury and disease modeling

Modeling a skin complex layered structure provided tissue-specific functionality is one of the actual challenges in Regenerative Medicine. In this study, we have utilized novel organoids (SO) containing key cell types as tool for vitro disease modeling and chemical irritation testing immersive conditions. Key human cells were induced to form SO further exposed UVB, retinol, 1% Triton solution, Isopropanol, Hexyl Salicylate, 5%KOH on d7. Melanoma added days 0 7 melanoma modeling. Formed analyzed by Live/Dead assay, histology, IHC, Photometry, RT-PCR, etc at 7, 14, & 21 cultivation. Statistics performed using Graphpad Software Inc. Skin self-organized into spherical with skin-like microstructure, surface zone formed epidermis central core dermal hypodermal cells. As well, showed epidermal barrier formation, vasculogenesis, pigmentation. Generated capable utilize retinol retinoic acid develop ER-stress reaction response UVB exposure, responded skin-irritating chemicals accordingly their index. The that malignant rapidly growing tumor spheroid outside organoid small cluster foci organoid's core. Therefore, study multicellular showing are high-throughput analysis conditions, able recapitulate well physiological response. Ultimately, technique provides an model could be used platform investigation dermatopathology.
